2,3,4,5-Tetra(4-pyridyl)thiophene

GANT 58 (chemical name  2,3,4,5-Tetra(4-pyridyl)thiophene) is a biochemical agent used in medical and pharmacological research as a Hedgehog signaling pathway inhibitor (specifically as a GLI1 antagonist).

The biological mechanismof GANT 58 is to block the Hedgehog signaling pathway relatively far downstream in the signaling cascade (downstream of Smoothened and Sufu). It specifically prevents gene transcription mediated by the GLI1 protein within the cell nucleus.

GANT 58 is being investigated in cancer research. In laboratory studies, it demonstrates antiproliferative and anti-tumor properties, for example, in models of Ewing sarcoma or acute T-cell leukemia.
